Bone Defect Repair

Bone defect repair is the restoration of damaged or missing bone to reestablish structural strength, function, and tissue continuity. Treatment may combine bone grafts, biomaterial scaffolds, fixation, and regenerative approaches that support cell attachment, vascularization, and new bone formation while the immune system coordinates inflammation and healing. In immunology and infection research, repair strategies are evaluated for how they regulate macrophage responses, limit bacterial colonization, and integrate with host tissue. Understanding these interactions can improve outcomes for trauma, surgery, chronic wounds, and implant-associated infections while guiding the development of safer, more effective regenerative therapies.

Here, we present a protocol to visualize blood vessel formation in vivo and in real-time in 3D scaffolds by multiphoton microscopy. Angiogenesis in genetically modified scaffolds was studied in a murine calvarial critical bone defect model. More new blood vessels were detected in the treatment group than in controls.

One constraint of preclinical research in the field of bone repair is the lack of experimental control over the local mechanical environment within a healing bone lesion. We report the design and use of an external fixator for bone repair with the ability to change fixator stiffness in vivo.
