Cd14+ Monocytes

CD14+ monocytes are circulating innate immune cells identified by surface expression of the CD14 co-receptor, which makes them important responders to infection, tissue injury, and inflammation. CD14 binds bacterial lipopolysaccharide together with LPS-binding protein and facilitates signaling through the TLR4 complex, triggering cytokine production, antimicrobial activity, and changes in gene expression; these cells can also differentiate into macrophages or dendritic cells after entering tissues. In immunology and infection research, flow cytometry distinguishes CD14+ monocyte subsets and tracks their abundance, activation, and maturation. Such analyses help characterize host responses, inflammatory disease, sepsis, and pathogen-associated immune dysfunction.

Osteoclasts are key bone-resorbing cells in the body. This protocol describes a reliable method for the in vitro differentiation of osteoclasts from human peripheral blood monocytes. This method can be used as an important tool to further understand osteoclast biology in homeostasis and in diseases.

This study compares two different methods of human monocyte isolation for obtaining in vitro dendritic cells (DCs). Monocytes are selected by adherence or negatively enriched by magnetic separation. Monocyte yield and viability along with MDDC viability, proliferation and CD11c/CD14 surface marker expression will be compared between both methods.

An In Vitro Assay for Evaluating the Immunomodulatory Impact of Monocytes on Leukocyte Proliferation

This video demonstrates an in vitro assay to explore the immunomodulatory influence of mesenchymal stem cells (MSCs) on effector T cell proliferation. The assay includes co-culturing MSCs with PBMCs to induce the monocyte population to acquire an immunomodulatory phenotype, leading to the secretion of interleukin-10 (IL-10). Following incubation with activated CD4+ T cells, the induced monocytes effectively suppress T cell proliferation.

We describe a procedure to assess the capacity for pharmacologic agents to generate tolerogenic dendritic cells from naïve monocyte-derived dendritic cells in vitro and validate their potency by autologous regulatory T cell generation.
