Cell Extract Preparation

Cell extract preparation is the process of disrupting cells and recovering their soluble intracellular contents for biochemical, immunological, or infection-related analysis. It typically combines mechanical or chemical lysis with controlled buffer conditions that stabilize target proteins, nucleic acids, or other molecules, followed by centrifugation or filtration to remove intact cells and debris. In immunology and infection research, these extracts can provide antigenic material, host-response factors, or pathogen-associated components for assays such as immunoblotting, enzyme activity measurements, and immune-recognition studies. Careful control of lysis strength, temperature, and protease activity improves extract quality and helps preserve biologically informative signals.

Crude and fractionated Xenopus egg extracts can be used to provide ingredients for reconstituting cellular processes for morphological and biochemical analysis. Egg lysis and differential centrifugation are used to prepare the crude extract which in turn in used to prepare fractionated extracts and light membrane preparations.

We describe a reliable method for the preparation of whole cell extracts from yeast or other cells using a cryogenic freezer mill that minimizes degradation and denaturation of proteins. The cell extracts are suitable for purification of functional protein complexes, proteomic analyses, co-immunoprecipitation studies and detection of labile protein modifications.
