Crude Extracellular Mixture

A crude extracellular mixture is an unpurified collection of molecules released by cells into the surrounding environment, often recovered from cell-free culture fluid. It may contain secreted proteins, enzymes, metabolites, membrane vesicles, and other soluble or particulate factors whose combined activity reflects the producing organism or cell population. In immunology and infection research, investigators expose immune cells or host tissues to these mixtures and measure outcomes such as cytokine production, inflammatory signaling, cytotoxicity, or antimicrobial responses. Although its components are not individually defined, the mixture can reveal the collective effects of extracellular products and guide later fractionation or identification of specific immunomodulatory factors.

The pH of a solution containing an acid can be determined using its acid dissociation constant and its initial concentration. If a solution contains two different acids, then its pH can be determined using one of several methods depending upon the relative strength of the acids and their dissociation constants. A Mixture of a Strong Acid and a Weak Acid In a mixture of a strong acid and a weak acid, the strong acid dissociates completely and becomes a source of almost all the hydronium ions...

Source: Wiegand, D. J., et al. Cell-free Protein Expression Using the Rapidly Growing Bacterium Vibrio natriegens. J. Vis. Exp. (2019).This video demonstrates the preparation of a cell extract from Vibrio natriegens, a rapidly growing bacterium known for its high protein synthesis rate. The bacterial pellet is washed and resuspended in lysis buffer before sonication on ice to prevent cellular degradation. The lysate is centrifuged to separate insoluble cell debris, and the cell extract is...

A multidimensional gas chromatography method for the analysis of dissolved hydrogen sulfide in liquid crude oil samples is presented. A Deans switch is used to heart-cut light sulfur gases for separation on a secondary column and detection on a sulfur chemiluminescence detector.

Separation of Mixtures via Precipitation

Source: Laboratory of Dr. Ana J. García-Sáez — University of Tübingen Most samples of interest are mixtures of many different components. Sample preparation, a key step in the analytical process, removes interferences that may affect the analysis. As such, developing separation techniques is an important endeavor not just in academia, but also in industry. One way to separate mixtures is to use their solubility properties. In this short paper, we will deal with aqueous solutions. The...
