Immunomagnetic Sorting

Immunomagnetic sorting is a cell-separation method that isolates specific cell populations from a mixed sample using antibody-coated magnetic particles. The antibodies bind surface markers on target cells, and applying a magnetic field retains either the labeled cells or the unlabeled fraction, depending on the selection strategy. This approach enables rapid enrichment or depletion of immune cells from blood, tissue, and other biological specimens while preserving cells for downstream analysis or culture. In immunology and infection research, immunomagnetic sorting supports pathogen-host studies, immune-cell profiling, diagnostic workflows, and the preparation of defined populations for functional assays.

We present the techniques required to isolate the stromal vascular fraction (SVF) from mouse inguinal (subcutaneous) and perigonadal (visceral) adipose tissue depots to assess their gene expression and collagenolytic activity. This method includes the enrichment of Sca1high adipose-derived stem cells (ASCs) using immunomagnetic cell separation.

Isolating Murine Primary Oligodendrocytes Using Immunomagnetic Beads

This video demonstrates a method to isolate primary oligodendrocyte cells from a neural cell suspension obtained from a mouse pup using magnetic separation. The technique employs magnetic beads coated with antibodies specific to oligodendrocyte antigens, enabling selective isolation of these cells.

Negative Immunomagnetic Selection: A Method to Purify B-cells from Peripheral Blood Mononuclear Cells

2023

The video explains a way to purify B-cells by immunomagnetic targeting and separation from peripheral blood mononuclear cells suspension. The method involve labeling unwanted cell types with specific antibodies or ligands (linked to magnetic particles) targeting specific cell surface proteins, to be depleted from the heterogenous cell suspension by magnetic separation.
