Immune Tolerance Studies

Immune tolerance studies examine how the immune system avoids attacking the body’s own tissues while remaining capable of responding to harmful foreign substances. This balance develops through central tolerance, which removes or alters self-reactive lymphocytes during immune-cell maturation, and peripheral tolerance, which suppresses escaped autoreactive cells through regulatory T cells, anergy, or immune regulation. In medicine, these studies clarify the origins of autoimmune disease, transplant rejection, and failures of immune tolerance. Their findings support strategies that promote antigen-specific tolerance, improve graft survival, and guide treatments designed to control harmful immune responses without broadly weakening protective immunity.

Herein we describe two approaches to characterize cell polarization events in B lymphocytes during the formation of an IS. The first, involves quantification of organelle recruitment and cytoskeleton rearrangements at the synaptic membrane. The second is a biochemical approach, to characterize changes in composition of the centrosome, which undergoes polarization to the immune synapse.
