Outflow Graft

An outflow graft is a surgically implanted conduit that directs blood from a cardiac chamber or mechanical circulatory support device into an artery, restoring or augmenting forward circulation when native cardiac output is inadequate. In left ventricular assist devices, the graft connects the pump outlet to the aorta, allowing blood drawn from the left ventricle to bypass weakened pumping function and reach systemic circulation. Its design, attachment, and patency are important because flow obstruction, kinking, or malposition can reduce support and cause complications. Surgeons and researchers use imaging, operative assessment, and device monitoring to evaluate graft function, guide treatment, and improve outcomes in advanced heart failure.

Marginal grafts, such as fatty livers, grafts from older donors, or livers retrieved after cardiac death (DCD) tolerate conventional, cold static storage only poorly. We developed a novel model of subnormothermic ex vivo liver perfusion for preservation, assessment, and repair of marginal liver grafts prior to transplantation.

The severe organ shortage has resulted in increased use of marginal kidney grafts for transplantation. This has triggered interest in alternative storage methods, since marginal grafts especially tolerate cold storage poorly. The technique of normothermic ex vivo kidney perfusion (NEVKP) represents a novel preservation method for kidney grafts prior to transplantation.

Mouse Models for Graft Arteriosclerosis

We describe protocols for our mouse graft arteriosclerois (GA) models which involve interposition of a mouse vessel segment into a recipient of the same inbred strain. By backcrossing additional genetic changes into the vessel donor, the model can assess the effect of specific genes on GA.

A Technique for Human Skin Grafting in a Mouse Model

This video demonstrates a method for grafting human skin onto an immunodeficient mouse with a suppressed neutrophil response. The method involves applying human split-thickness skin to the mouse's graft bed, sealing it, and covering it for recovery. Eventually, the human skin adheres to the mouse's skin, establishing vascularization and circulation, resulting in a successful skin xenograft.

This report provides a detailed description of transplanting murine thymi from different aged donor mice under the kidney capsule of immunodeficient mouse recipients. The goal of this approach is to model T cell development and thymic selection events in vivo.
