Activity Tube Preparation

Activity tube preparation is the standardized assembly of tubes used to measure locomotor activity in animal models, providing a controlled setting for behavioral neuroscience experiments. Tubes are typically fitted with an appropriate food or support medium, sealed to maintain the test environment, and positioned in an activity monitor where movement is detected as interruptions of an infrared beam. Consistent tube dimensions, loading conditions, and environmental controls improve the reliability of activity counts over time. This preparation supports studies of locomotion, sleep-wake behavior, circadian rhythms, and neurological phenotypes, enabling quantitative comparisons of behavior across experimental groups.

We describe a process for fixation, embedding, sectioning, and imaging of late stage Drosophila embryos for Trasmission Electron Microscopy of the embryonic heart tube.
