Butterfly Eye Preparation

Butterfly eye preparation is a laboratory method for isolating and maintaining the butterfly visual system so researchers can examine how light is detected and encoded by neural tissue. The procedure preserves the retina and connections to the optic lobe, allowing investigators to stimulate the eye with controlled light and record responses from photoreceptors or downstream neurons. Because butterfly eyes contain specialized visual adaptations, this preparation supports studies of phototransduction, neural signaling, and visual circuit organization. In situ Protocol for Butterfly Pupal Wings Using Riboprobes

In order to examine gene expression in the pupal wing tissue of Bicyclus anynana, we present an optimized protocol for in situ hybridizations using riboprobes. We also provide guidelines for the further optimization of this protocol for use in pupal wings of other Lepidopteran species.

Preparation of Adult Drosophila Eyes for Thin Sectioning and Microscopic Analysis

A standard approach to prepare adult Drosophila eyes for semi-thin sectioning and light microscopic analysis is presented here. The protocol can be used for gross morphological analysis of eye defects, or with the indicated adjustments can be used to determine genetic requirements of genes in specific cell types of the eye (e.g. clonal analysis of photoreceptors) or for electron microscopic analysis.
