Calcium Measurement

Calcium measurement is the quantitative assessment of calcium ion concentration or changes in calcium levels within cells, tissues, or extracellular environments. In neuroscience, fluorescent calcium indicators bind Ca2+ and alter their fluorescence, allowing microscopes to convert activity-dependent calcium influx into measurable signals; genetically encoded indicators can be targeted to specific neurons or compartments. These measurements reveal patterns of neuronal firing, synaptic activity, intracellular signaling, and network dynamics, complementing electrophysiological recordings. Calcium imaging is therefore widely used to study brain circuits, sensory processing, learning, disease mechanisms, and responses to pharmacological or genetic manipulation.

Measuring Fast Calcium Fluxes in Cardiomyocytes

We present a method to isolate rapid (microsecond) calcium events from slower fluxes in living cells using laser scanning confocal microscopy. The method measures fluorescence intensity fluctuations of calcium indicators by recording line scans of several hundred pixels in a cell. Histogram analysis allows us to isolate the time scales of different calcium fluxes.
