Capillary Network Analysis

Capillary Network Analysis is the quantitative study of the structure and organization of small blood-vessel networks, including their density, branching, length, diameter, and connectivity. In neuroscience, it typically uses microscopy or vascular imaging followed by image processing to segment capillaries and calculate network features that describe how blood vessels are arranged within brain tissue. These measurements help relate microvascular architecture to oxygen and nutrient delivery, neurovascular interactions, development, and changes associated with neurological disease. Comparing network properties across regions or experimental conditions can reveal vascular remodeling and clarify how blood supply influences neuronal function.

Capillary Electrophoresis (CE)

2023

Source: Laboratory of Dr. B. Jill Venton - University of Virginia Capillary electrophoresis (CE) is a separation technique that separates molecules in an electric field according to size and charge. CE is performed in a small glass tube called a capillary that is filled with an electrolyte solution. Analytes are separated due to differences in electrophoretic mobility, which varies with charge, solvent viscosity, and size. Traditional electrophoresis in gels is limited in the amount of voltage...

This video demonstrates the isolation of capillaries from fresh porcine brains. The process involves removing the meninges, isolating the grey matter from the brain tissue, homogenizing and filtering the grey matter to isolate the capillaries, and finally using enzymatic digestion to purify the capillaries further.

This protocol combines the characterization of a protein sample by capillary gel electrophoresis and a fast-binding screening for charged ligands by affinity capillary electrophoresis. It is recommended for proteins with a flexible structure, such as intrinsically disordered proteins, to determine any differences in binding for different conformers.
