Drug Delivery Enhancement

Drug delivery enhancement refers to strategies that improve a therapeutic agent’s transport, stability, targeting, or release so it reaches its intended site at an effective dose. In neuroscience, these strategies address barriers such as the blood–brain barrier through nanoparticle carriers, chemical modification, receptor-mediated transport, or intranasal administration. For example, a drug linked to a receptor-binding ligand can trigger transcytosis across brain endothelial cells, while controlled-release formulations protect drugs from degradation and maintain exposure. These approaches support research and treatment development for neurological disorders, but their safety, distribution within neural tissue, and therapeutic efficacy require careful evaluation.

Take cultures of human monocyte-derived microglia-like cells.Introduce nanoparticles containing an anti-inflammatory drug into the experimental culture and the free drug into the control. The nanoparticles feature a hydrophilic shell and a hydrophobic core encapsulating the lipophilic drug. The cells endocytose the nanoparticles, which release the drug intracellularly, enhancing its delivery. The free drug enters by passive diffusion, a process constrained by the concentration gradient, which...

Begin with an anesthetized human patient diagnosed with recurrent glioblastoma, an invasive tumor characterized by tumor cell infiltration into the surrounding peritumoral region. The peritumoral region retains an intact blood-brain barrier (BBB), where tight junctions between endothelial cells restrict drug penetration.Make an incision in the scalp, create an opening in the skull, and insert a laser probe into the tumor core using MRI guidance.Deliver laser light through the probe.Tissue...

Convection-Enhanced Delivery of Antibodies into a Mouse Brain

This video demonstrates a convection-enhanced delivery (CED) technique for antibody delivery into the mouse brain. The skull of an anesthetized mouse is exposed, and a burr hole is created at the target area for antibody delivery. A step catheter containing antibodies is inserted at the target site, and via an infusion pump, a pressure gradient is created to deliver antibodies at a controlled rate.

Source: Kanazawa, T., et al. Novel Methods for Intranasal Administration Under Inhalation Anesthesia to Evaluate Nose-to-Brain Drug Delivery. J. Vis. Exp. (2018)This video demonstrates the intranasal administration of radiolabeled drugs in an anesthetized mouse to achieve nose-to-brain drug delivery. It explains how drugs cross the nasal epithelium and travel via neuronal pathways to reach the brain, as well as how ciliary motion influences drug retention and clearance. Additionally, the video...
