Focal Brain Injury

Focal brain injury is damage confined primarily to a specific region of the brain, often producing neurological deficits that reflect the affected area. It can result from trauma, stroke, tumors, or infection, and may disrupt neuronal circuits through tissue loss, bleeding, swelling, and inflammation. In neuroscience, examining the relationship between lesion location and changes in movement, sensation, language, or memory helps clarify how brain regions support behavior. Clinical imaging and neurological assessment guide diagnosis and treatment, while focal injury models support research on neural plasticity, functional recovery, and rehabilitation strategies.

This protocol describes a custom overpressure air system designed to induce closed-system central nervous system (CNS) injuries in mice, including ocular, brain, and spinal cord traumas. Athletes absorb several hundred mild traumatic brain injuries (mTBI)/concussions every year; however, the consequence of these on the brain is poorly understood. Therefore, an animal model of single and repetitive mTBI that consistently replicates clinically relevant symptoms provides the means to advance the study of mTBI and concussion.

Lateral Fluid Percussion: Model of Traumatic Brain Injury in Mice

Lateral fluid percussion (LFP), an established model of traumatic brain injury in mice, is demonstrated. LFP fulfills three major criteria for animal models: validity, reliability and clinical relevance. The procedure, consisting of surgical craniotomy, fixation of hub followed by induction of injury, resulting in focal and diffuse injuries, is described.
