Nerve Cord Isolation

Nerve cord isolation is the dissection-based separation of neural tissue from surrounding structures, enabling researchers to study the organization and function of the central nervous system in a controlled preparation. Under microscopic guidance, surrounding tissues are carefully removed while the nerve cord remains intact and is maintained in an appropriate physiological solution to preserve cellular activity and anatomical relationships. In neuroscience, isolated nerve cords support electrophysiological recording, neural circuit analysis, imaging, molecular studies, and ex vivo culture. This preparation helps link neuronal structure with sensory processing, motor output, and network activity while reducing the complexity of the intact organism.

Combining Peripheral Nerve Grafting and Matrix Modulation to Repair the Injured Rat Spinal Cord

Traumatic injury to the spinal cord disrupts communication with the brain. To restore lost connectivity we utilize a peripheral nerve graft to provide a substratum for regenerating fibers in combination with neurotrophic factors and matrix-modulating enzymes to remove inhibitory molecules to promote long distance growth.

Hydraulic Extrusion of the Spinal Cord and Isolation of Dorsal Root Ganglia in Rodents

Here, we present a protocol for hydraulic extrusion of the spinal cord as well as identification and isolation of specific dorsal root ganglia (DRGs) in the same rodent. Compared to standard spinal cord isolation methods, this method is significantly faster and reduces the risk of tissue damage.

We demonstrate a protocol for axoplasm isolation from adult rat sciatic nerve based on dissection of nerve fascicles and incubation in hypotonic medium to release myelin and lyse non-axonal structures, followed by extraction of the remaining axon-enriched material.

Whole Mount Preparation of the Adult Drosophila Ventral Nerve Cord for Giant Fiber Dye Injection

An in vivo dissection of the adult Drosophila ventral nerve cord (VNC) is demonstrated. This particular dissection method causes little damage to the VNC allowing the subsequent labeling of the giant fiber neurons with fluorescent dye for high resolution imaging.
