Olfactory Epithelium Stimulation

Olfactory epithelium stimulation is the activation of sensory neurons within the specialized nasal tissue that detects airborne chemicals, providing the first step in odor perception. When odorant molecules dissolve in the mucus layer and bind to receptors on olfactory sensory neuron cilia, they trigger a G protein-mediated signaling cascade that increases intracellular cyclic AMP, opens ion channels, and generates electrical signals. In neuroscience, controlled stimulation helps researchers examine odor coding, sensory transduction, and neural pathways from the nose to the olfactory bulb and higher brain regions. These approaches support studies of perception, behavior, respiratory influences on sensing, and disorders that affect smell.

Navigational Behavior of Drosophila Larvae in Response to Optogenetic Stimulation of Olfactory Sensory Neurons

2025

This video demonstrates a study of the navigational behavior of Drosophila larvae in response to optogenetic stimulation of olfactory sensory neurons (ORNs). Transgenic Drosophila larvae expressing light-sensitive ion channels in the ORNs are placed on agarose within a behavior arena. Optogenetic stimulation with light opens the channels, causing a cation influx that triggers the transmission of the signal to the brain, and the navigation behavior is recorded.

Cilia of olfactory sensory neurons contain proteins of the signal transduction cascade, but a detailed spatial analysis of their distribution is difficult in cryosections. We describe here an optimized approach for whole mount labeling and en face visualization of ciliary proteins.

Perforated Patch-Clamp Recording of Olfactory Sensory Neurons in Mice

2025

This video demonstrates electrophysiological recording of olfactory sensory neurons (OSNs) in mice using a perforated patch-clamp technique. The olfactory epithelium, containing OSNs with fluorophore-tagged odor receptors, is isolated from a mouse and secured in a recording chamber. A recording pipette is clamped to the dendritic knob of an OSN, and nystatin within the pipette perforates the membrane to form electrical connectivity with the cell. Finally, another pipette is used to deliver an...
