Osmium Ferrocyanide

Osmium ferrocyanide is a specialized electron-dense staining and postfixation reagent used to preserve and visualize lipid-rich biological membranes, making it valuable for examining neural ultrastructure. In practice, potassium ferrocyanide reduces osmium tetroxide during fixation, promoting deposition of reduced osmium in membranes such as myelin and synaptic structures. This enhanced contrast supports electron microscopy of axons, dendrites, synapses, and organelles while helping maintain structural detail in prepared tissue. In neuroscience, the method aids studies of connectivity, myelination, and neurodegenerative changes, where membrane integrity and ultrastructural organization are important.
