Uranyl Acetate Staining

Uranyl acetate staining is an electron microscopy technique that enhances contrast in biological specimens, allowing fine cellular structures to be visualized at high resolution. The uranyl-containing stain binds preferentially to electron-dense or negatively charged components, and its high atomic number scatters electrons strongly during transmission electron microscopy. In neuroscience, the method helps reveal neuronal membranes, synaptic vesicles, organelles, and other ultrastructural features in fixed, dehydrated tissue sections. By improving structural contrast, uranyl acetate staining supports analysis of synaptic organization, neurodegenerative changes, and cellular responses to experimental conditions.

A method for the determination of acetate kinase activity is described. This assay utilizes a direct reaction for determining enzyme activity and kinetics of acetate kinase in the acetate-forming direction with different phosphoryl acceptors. Furthermore, this method can be utilized for assaying other acetyl phosphate or acetyl-CoA utilizing enzymes.
