Oxidative Current Measurement

Oxidative current measurement is an electrochemical technique that detects electroactive molecules by measuring the electrical current produced during their oxidation at an electrode. In a typical experiment, a controlled potential drives electron transfer from molecules such as dopamine to the electrode, and the resulting current reflects the amount reaching its surface over time. In neuroscience, this approach can monitor rapid neurotransmitter release in brain tissue, helping researchers relate chemical signaling to neuronal activity and circuit function. Its temporal and chemical sensitivity supports studies of synaptic transmission, neuromodulation, and disease-related changes, while electrode design and calibration determine measurement specificity and accuracy.

Comet assay measures DNA breaks, induced by different factors. If all factors (except oxidative stress) causing DNA damage are kept constant, the amount of DNA damage is a good indirect parameter of oxidative stress. The goal of this protocol is to use comet assay for indirect measurement of oxidative stress.

This video demonstrates a quantification technique for CO2 released during substrate oxidation using 14C-radiolabeled substrates. The released 14CO2 is trapped in an alkaline solution and quantified by a scintillation counter. The oxidation of different substrates varies between tissues and reflects the pathophysiological condition of the tissue.
