Oxygenated Saline

Oxygenated saline is an aqueous solution enriched with dissolved oxygen to support the metabolic needs of isolated or excised biological tissues. In neuroscience, it is commonly prepared by bubbling saline with an oxygen-containing gas, allowing oxygen to diffuse through the solution while its ions, pH, and osmolarity maintain a controlled extracellular environment. This solution helps preserve neuronal function in acute brain slices, tissue preparations, and perfusion-based experiments, where oxygen delivery is limited after blood circulation stops. By sustaining cellular metabolism and physiological conditions, oxygenated saline enables electrophysiological recordings, synaptic studies, and investigations of neural circuits outside the intact organism.

Cerebral Blood Oxygenation Measurement Based on Oxygen-dependent Quenching of Phosphorescence

We present an experimental procedure for measuring the partial pressure of oxygen (pO2) in cerebral vasculature based on oxygen-dependent quenching of phosphorescence. Animal preparation and imaging procedures were outlined for both large field of view CCD-based imaging of pO2 in rats and 2-photon excitation based imaging of pO2 in mice.

A Hypertonic Saline Injection Technique for Inducing Glaucoma in Rats

This video demonstrates glaucoma induction in an anesthetized rat by injecting hypertonic saline into the episcleral vein. The saline enters the trabecular meshwork via Schlemm's canal, causing tissue scarring, which blocks aqueous humor outflow and raises intraocular pressure. The elevated pressure damages retinal ganglion cells, leading to glaucoma.
