Peptide Organization

Peptide organization is the structural and spatial arrangement of peptide molecules, from their amino acid sequences and three-dimensional conformations to their assembly within cells, and it is essential for understanding biological function. Organization arises through peptide folding, hydrogen bonding, electrostatic forces, hydrophobic interactions, and association with membranes or other molecules, while cellular trafficking determines where signaling peptides accumulate and act. In neuroscience, this framework helps explain how neuropeptides are packaged, released, and positioned to regulate neuronal communication, receptor signaling, and behavior. Studying peptide organization also supports research on protein aggregation, neurodegenerative disease, therapeutic design, and biomolecular engineering.

Personalized Peptide Arrays for Detection of HLA Alloantibodies in Organ Transplantation

Mismatches in human leukocyte antigen (HLA) sequences between organ donor and recipient pairs are the major cause of antibody-mediated rejection in organ transplantation. Here we present the use of custom antigen arrays that are based on individual donors' HLA sequences to probe anti-donor HLA alloantibodies in organ recipients.

This video demonstrates an assay to detect anti-donor HLA alloantibodies in organ recipients. A peptide array is synthesized to represent potential epitopes for antibody-mediated organ rejection. The array is incubated with the recipient's serum, containing alloantibodies that bind to their target epitopes. Antibody binding is detected using chemiluminescence to identify donor-specific HLA epitopes that incite alloantibody reactivity.
