Retinol Fluorescence

Retinol fluorescence is the emission of light by retinol, a vitamin A-derived molecule, after it absorbs higher-energy radiation. Its conjugated polyene structure absorbs ultraviolet or blue light, and the excited molecule releases part of that energy as longer-wavelength fluorescence; the signal can vary with solvent, molecular binding, and local chemical environment. In neuroscience, this property is relevant to the visual cycle, in which photoreceptors convert light and regenerate light-sensitive chromophore from vitamin A derivatives. Measuring retinol fluorescence can therefore help investigate retinoid metabolism, photoreceptor function, and changes in retinal tissue associated with visual disorders.

A method is described for the preparation of single living photoreceptor cells from different vertebrate species for fluorescence imaging. The method can be used to image the fluorescence of endogenous fluorophores, such as NADH or vitamin A, or that of exogenously added fluorescent dyes sensitive to Ca2+ or other factors.
