Retrograde Tracer

A retrograde tracer is a labeling substance used to identify neurons that project their axons to a specific region, making it an important tool for mapping neural connectivity. After injection into a target area, the tracer is taken up by axon terminals and transported backward along the axon to the neuronal cell body, where it accumulates for detection. Researchers use retrograde tracers to define pathways, characterize projection patterns, and examine the organization of brain circuits. Combined with microscopy or molecular markers, this approach links anatomical connections with neuronal identity and function in neuroscience research.

The efficacy of intramuscular uptake and retrograde transport of molecules to corresponding motor neurons depends on the location of the injection sites with respect to the motor end plates (MEPs). Here, we describe how to locate MEPs on skeletal muscles to optimise retrograde transport of tracers into motor neurons.

In order to study brain reorganization under pathological conditions we used miniosmotic pumps for direct protein delivery into the brain circumventing the blood brain barrier. Tract tracers are then injected to study alterations in brain connectivity under the influence of the protein.

Source: Inal, M. A. et. al., Retrograde Tracing of Drosophila Embryonic Motor Neurons Using Lipophilic Fluorescent Dyes. J. Vis. Exp. (2020)This video demonstrates the method of retrograde labeling of Drosophila motor neurons using lipophilic dyes. The dye enters the axon, travels retrogradely, and labels the neurons green, allowing detailed visualization of their morphology and projections under a confocal microscope.
