Vesicle Organization

Vesicle organization is the spatial arrangement, movement, and regulated clustering of membrane-bound compartments within cells and tissues. In neurons, this organization positions synaptic vesicles near active zones while trafficking pathways deliver vesicles to axons, dendrites, or recycling sites. Cytoskeletal networks, motor proteins, membrane-tethering factors, and SNARE-associated machinery coordinate vesicle transport, docking, fusion, and retrieval, enabling neurotransmitter release and membrane maintenance. Studying vesicle organization helps explain how synapses develop, adapt, and transmit signals, supporting research into neuronal communication, synaptic plasticity, and neurological disease.

We describe how to implement photoactivated localization microscopy (PALM)-based studies of vesicles in fixed, cultured neurons. Key components of our protocol include labeling vesicles with photoconvertible chimeras, collecting sparsely sampled raw images with a super-resolution microscopy system, and processing the raw images to produce a super-resolution image.
