<li>Solutions should be kept RNase-free. Use either glass-ware that has been baked (4 hours at 250&deg;C) or disposable plastic. Serological pipettes are very useful for making up solutions </li>

<li>Dissect wings from time-staged pupae in PBS</li>
<li>Move wings directly to Fix Buffer in wells of 24 well culture plate at room temperature</li>
<li>Fix discs for 2 hours </li>
<li>Wash 5 x 5 minutes in PBT</li>
<li>Incubate wings for 3 minutes in Proteinase K solution</li>
<li>Rinse 2 x in Digestion Stop Buffer</li>
<li>Wash 5 x 5 minutes in PBT</li>

<li>Wash 1 x 10 minutes in PHB</li>

<li>Develop wings in 1 ml of Developing Solution - time of development must be monitored each time - developing times can change even when using the same probes and developing solution - generally check after 5-10 minutes and then increase intervals up to overnight. Plate should be wrapped in foil to prevent light exposure when not checking samples.</li>
<li>Rinse five times in Developing Stop Buffer</li>

In situ Protocol for Butterfly Pupal Wings Using Riboprobes

Here we present, in video format, a protocol for in situ hybridizations in pupal wings of the butterfly Bicyclus anynana using riboprobes.  In situ hybridizations, a mainstay of developmental biology, are useful to study the spatial and temporal patterns of gene expression in developing tissues at the level of transcription. If antibodies that target the protein products of gene transcription have not yet been developed, and/or there are multiple gene copies of a particular protein in the genome that cannot be differentiated using available antibodies, in situs can be used instead. While an in situ technique for larval wing discs has been available to the butterfly community for several years, the current protocol has been optimized for the larger and more fragile pupal wings.

In order to examine gene expression in the pupal wing tissue of Bicyclus anynana, we present an optimized protocol for in situ hybridizations using riboprobes. We also provide guidelines for the further optimization of this protocol for use in pupal wings of other Lepidopteran species.

This is an ceto protocol for butterfly wings at the pupil stage. So we start by collecting pre pupa from the cage, setting them on a tray, and then photographing their exact PPU patient times by using time lapse photography. This allows us to do the stainings at precise times after pupation occurs.This is important because different mRNAs for different proteins are expressed at different times after pation. Here we're dissecting the pupil wings in a Petri dish with a silicone base, and the important part is to remove the wings and put them in the fix solution so that they are very flat. If they're not flat at this stage, they will never become flat again.After the right fixation time, we remove the fixed buffer and we wash five times. With PBT, the proteinase case step is rather critical, and it's used to per Alize the tissues for the probe to it should be stopped at the right time with these rinses and washes. The salmon sperm works as a cheap source of DNA to prevent the probe from binding everywhere.The sperm is one of the components of the pre hybridization buffer. So here we are adding the sperm to the pre hybridization buffer, and then there's going to be a solution made, whi
