This article discusses the application of deep transcranial magnetic stimulation (dTMS) in treating post-traumatic stress disorder (PTSD). It highlights the mechanism by which dTMS activates the medial prefrontal cortex (mPFC) to inhibit the overactive amygdala, thereby alleviating fear responses associated with PTSD.
Deep transcranial magnetic stimulation (dTMS) offers a non-invasive neuromodulation approach to target neural circuit dysfunction in PTSD, addressing a critical gap in CNS drug development where mechanistic de-risking is essential. By restoring prefrontal cortical control over limbic hyperactivity, dTMS provides a translatable biomarker-driven strategy for target validation in neuropsychiatric disorders. This supports early discovery efforts focused on circuit-based therapeutics and predictive confidence in target engagement.
The method fits within the discovery continuum from target validation through lead identification, where circuit modulation serves as a functional readout for compound screening and target modulation assessment.
