This study investigates the survival mechanisms of Mycobacterium tuberculosis under iron-limited conditions. It highlights the role of siderophores in iron acquisition and bacterial adaptation.
Understanding bacterial adaptation to iron limitation provides mechanistic insights into pathogen survival strategies, supporting target validation in antimicrobial discovery. This model enables phenotypic screening of compounds that disrupt siderophore-mediated iron acquisition, a validated virulence pathway. Mechanistic de-risking of anti-infective candidates is enhanced by linking target engagement to bacterial stress response pathways under physiologically relevant conditions.
This method positions iron-limited culture as a discovery-stage tool for target validation, feeding into lead identification pipelines focused on virulence mechanisms rather than essential growth.
