This article describes a protocol for differentiating human neural progenitor cells (NPCs) into mature neurons using a co-culture system with mouse astrocytes and rat cortical neurons. The NPCs are engineered to express a fluorescent protein for visualization under a confocal microscope.
Differentiating human neural progenitor cells into functional neurons is a critical step in target validation for neurodegenerative disease models. This co-culture technique enhances predictive confidence by providing a disease-relevant system that supports mechanistic de-risking of therapeutic hypotheses. The approach enables scalable preparation of validated neuronal phenotypes for downstream assay development and screening campaigns.
The method fits within the discovery continuum from stem cell-derived NPC generation to functional neuronal assays, supporting lead identification through phenotypic screening of compound effects on maturation and connectivity.
