This article describes a method for studying the effects of cholesterol synthesis inhibition on zebrafish embryos. The process involves dechorionating embryos and treating them with atorvastatin to induce hemorrhagic conditions for observation.
This method enables mechanistic de-risking of vascular toxicity by modeling drug-induced hemorrhage in a vertebrate system. It supports target validation and phenotypic screening for compounds affecting blood-brain barrier integrity. The assay provides quantitative, phenotype-based readouts for early safety assessment in discovery pipelines.
The assay fits within early discovery to evaluate target-mediated vascular toxicity before lead identification and preclinical development.
