This study investigates the effects of hemiparkinsonism on motor control in rats using a two-compartment staircase box. The experiment assesses the preference for paw usage in response to food pellets, highlighting the impact of dopamine neuron lesions in the right hemisphere of the brain.
The staircase test provides a quantitative behavioral readout for unilateral motor impairment in preclinical Parkinson's models, enabling target validation through functional assessment of dopaminergic pathway integrity. By measuring paw preference and pellet consumption asymmetry, the assay supports mechanistic de-risking of therapeutic candidates aimed at restoring motor symmetry. This approach enhances predictive confidence in early discovery by linking lesion-induced neurochemical deficits to observable functional outcomes.
The staircase test fits within the discovery continuum from target hypothesis testing to lead optimization, providing a functional motor endpoint that precedes preclinical efficacy studies.
