注意：稳定转染绿色荧光蛋白（GFP）和萤火虫荧光素酶基因（Fluc）的人类间充质干细胞（MSCs）购自商业公司（材料表）。
1. 表达萤火虫荧光素酶和绿色荧光蛋白的人类间充质干细胞的表征
<ol>
	<li>在完全培养基（最小必需培养基 eagle-α 改良型 [αMEM]，添加 10−15% 胎牛血清 [FBS]、2 mM L-谷氨酰胺、1 ng/mL 碱性成纤维细胞生长因子 [bFGF] 以及青霉素-链霉素-两性霉素 B 混合液 [PSF]）中，于 37 °C、饱和湿度及 5% CO2 条件下培养转染了 GFP 和 Fluc 的人源 MSCs。当细胞融合度达到约 70−90% 时进行传代。</li>
	<li>在荧光相差显微镜下观察 MSCs（图 1A），并分析 Fluc 和 GFP 的表达水平14。</li>
	<li>通过流式细胞术分析 CD44、CD73、CD90 和 CD105 等表面标志物的表达，对 MSCs 进行表型鉴定（图 1B）。诱导干细胞向脂肪细胞、软骨细胞和成骨细胞三系分化，并依据商业试剂盒说明书，通过 von Kossa 染色、油红 O 染色和阿利新蓝染色确认三系分化能力（图 1C）15,16。</li>
</ol>
2. 大鼠幼崽的麻醉
<ol>
	<li>让经时间标记的怀孕Sprague–Dawley大鼠在足月时经阴道自然分娩。</li>
	<li>在出生后第5天将大鼠幼崽从笼中取出。</li>
	<li>使用气体麻醉剂（例如，2%异氟烷）在麻醉舱中对大鼠幼崽进行麻醉。</li>
	<li>通过检查呼吸和反射确认麻醉充分。 
	注意：呼吸应变得浅慢，反射应减弱。在此异氟烷浓度下，大鼠幼崽至少保持无意识状态约10分钟。</li>
</ol>
3. 气管内灌注
<ol>
	<li>麻醉后，将幼鼠固定在倾斜约60°的插管架上，并用实验室标签胶带固定四肢。</li>
	<li>在鼻部下方贴胶带以固定头部，并确定颈部穿刺气管切开的位置。</li>
	<li>用75%酒精消毒片对切口区域（即颈部）进行消毒。</li>
	<li>使用显微剪刀在气管上方做一条0.3 cm的垂直正中颈部切口，注意避免损伤颈动脉。</li>
	<li>用无钩的尖头弯镊分离脂肪和肌肉层，以暴露气管。</li>
	<li>用尖头弯镊夹住气管。</li>
	<li>手持100 μL注射器直立，于吸气相通过30 G针头向气管内缓慢注入30 μL生理盐水（即对照组）或30 μL Fluc-GFP标记的MSCs（1 × 105 个细胞）。</li>
	<li>用一根6-0丝线缝合切口，打结尽量小，并将线头剪至最短。</li>
	<li>将大鼠置于红外灯下或加热垫上保温，使其从麻醉中恢复。</li>
	<li>确认大鼠体温正常、肤色粉红且能自主活动后，将其放回笼中。</li>
</ol>
4. 监测间充质干细胞在肺部的分布

	<li>为追踪移植的人源MSCs，在MSC注射后15分钟，以每公斤体重125 mg的剂量，通过腹腔注射将磷酸盐缓冲液（PBS）中的萤光素钾盐注入大鼠体内。</li>

新生儿呼吸窘迫常需补充氧气进行治疗1。然而，对婴儿实施高氧治疗会带来不良的长期影响。长时间暴露于高浓度氧气可导致炎症和急性肺损伤，其表现类似于人类支气管肺发育不良（BPD）2。尽管已采用早期表面活性物质治疗、优化通气策略以及在早产儿中增加无创正压通气的应用，BPD 仍是高氧治疗的主要并发症之一。虽然已有多种针对 BPD 的治疗策略被报道3，但目前尚无已知疗法能够有效减少该并发症的发生。

Intratracheal Instillation of Stem Cells in Term Neonatal Rats

气管内灌注是一种临床生物技术，用于将干细胞和药物递送至新生肺组织，以评估其治疗效果。该技术在新生大鼠中具有多种主要应用，是一种相对简便且经济有效的肺部疾病治疗方法。在出生后第5天，将麻醉后的Sprague-Dawley幼鼠固定于60度角的插管架上。将实验室标签胶带固定在每只幼崽的所有四个肢体上，以将其稳妥固定。在每只幼崽鼻部下方贴上胶带以固定头部。确定第一颈椎位置，以便进行气管穿刺。使用75%酒精消毒片对切口区域进行消毒。在气管上方沿中线做一条3厘米长的垂直切口，以避免损伤颈动脉。使用无钩的弯头尖镊子分离脂肪层和肌肉层，以定位气管。当观察到气管时，用镊子夹住气管，并使用配备30号针头的100微升注射器，在吸气相将30微升生理盐水中含1×10⁵个萤光素酶-GFP标记的间充质基质细胞注入气管。待细胞全部注入后，用160丝线缝合切口，尽可能扎紧结，缝线末端剪至尽可能短。然后将幼鼠置于温暖处，在密切监测下使其从麻醉中恢复，直至幼鼠体温恢复正常、肤色红润并能自主活动，再将其放回笼中。为追踪移植的人源间充质基质细胞，在大鼠术后恢复15分钟后，向重新麻醉的幼鼠腹腔注射125毫克/千克的荧光素钾盐（Luciferin potassium salt）PBS溶液。注射后10分钟，使用小动物成像系统进行连续成像，采集参数设置为中等 binning 模式、光圈f-stop值为1、视野直径26厘米，图像采集间隔为5至15秒。然后使用成像软件根据自动划定的兴趣区域定量分析肺部的发光活性。在荧光素酶标记的人源间充质干细胞中可观察到高水平的GFP表达。间充质干细胞还可通过其细胞表面标志物的表达以及分化为成骨细胞、软骨细胞和脂肪细胞的能力进一步鉴定。体内移植的人类间充质细胞的发光成像显示，经生理盐水处理的大鼠肺部区域无发光信号。而在经间充质干细胞处理的大鼠中，可在气管及中央肺区观察到发光信号。事实上，在此代表性分析中，发光强度的定量结果显示，与生理盐水处理组大鼠相比，间充质干细胞处理组大鼠的发光活性约增加了13倍。进行该操作时，重要的是在不损伤周围动脉的情况下解剖并分离气管。该技术可用于研究通过气管内给药方式向新生小鼠Palmer病模型施用细胞和药物的效果，其模式模拟人类患者的治疗情况。
