منشطات النسخ هى بروتينات مسؤولة عن السماح آر إن أيه بوليميراز ببدء عملية النسخ. لهم نطاقين أساسيين, واحد يرتبط ب دى إن أيه والآخر الذي ينشط النسخ. نطاقات ال دى إن أيه إحدى الخصائص التركيبية الزخرفية التسلسل المتعددة بما في ذلك زخارف تسلسل شائعة،مثل أشكال لفة الحلزون الحلزوني إصبع الزنك،السحاب ليوسين.النطاق فى الشكل الولبي helix-turn-helix مكون من حلزونين الفا متحدين مع بعض بواسطة سلسلة حمض أميني الذي يمثل اللّفة. أحد الحلزونين هو المسؤول عن التعرف على تسلسل دى إن أيه ويتوافق مع الأخدود(الحز)الرئيسي. هذه البروتينات ترتبط ب دى إن أيه كثنائى الوحدات.يحتوي إصبع الزنك على حلزون ألفا وشريطان بيتا متماسكة مع بعضها عن طريق ذرة زنك يرتبط اللولب مع الأخدود الرئيسي بمساعدة اثنين من الهيستيدين وسلسلة الحمض الأميني الخاص به. السحّاب ليوسين مكون من وحدات ثنائية التى تتفاعل مع بعضها البعض على شكل واى التى يرتبط ب دى إن أيه. تتكون أحادى الوحدات من ليوسين في كل موضع سابع في طرف النهاية الكربوكسيلية للحمض الأمينى،التي تتفاعل مع الليوسين على أحادى الوحدات المقابلة.الطرف النهائى N الخاص بأحادى الوحدات،الذى يتكون من زخارف تسلسلية بسيطة،ترتبط ب دى إن أيه. النطاق الأساسي الآخر في المنشط،نطاق تنشيط النسخ, توظّف البروتينات الأساسية المنشطة المساعدة،التى تتوسط في ربط المنشّطات ب آر إن أيه. هذه المنشطات المساعدة تحفز النسخ من خلال عدة آليات،مثل تعديل الهيستون،الذى يؤدي إلى زيادة إمكانية الوصول إلى آلية نسخ إلى دى إن أيه.يمكن للمنشطات أيضًا إرسال الإشارات عبر منشطات المساعدة الخاصة بهم لتنشيط آر إن أيه بوليميراز وبدء النسخ. في بعض الحالات،تكون المنشّطات أساسية في إستطالة النسخة. بعض البوليميرازيس توقف النسخ بعد قليل من النيوكليوتيدات وتحتاج لوجود منشط لإعادة بدء النسخ.نسخ جين واحد يمكن أن يخضع للتنظيم من قبل العديد من منشطات مختلفة. عندما يشترك أكثر من منشط في تنظيم النسخ, بوسعهم أن يتصرفوا على نحو متآزري لزيادة كبيرة فى معدل النسخ مقارنة ب المنشطات الفردية.

Transcription activators are proteins that help turn genes on by promoting transcription from DNA to RNA. They often have two parts. One part binds to DNA, and the other part activates transcription. In some proteins, such as the glucocorticoid receptor and MyoD, one domain can do both jobs.

The DNA-binding part recognizes specific regulatory sequences on the DNA. These binding domains are grouped into families based on their structure. Common examples include the leucine zipper, zinc finger, and helix-turn-helix motifs. The activation part is usually shorter and simpler. It is often described by amino acid content, such as glutamine-rich, proline-rich, or alanine-rich sequences.

Transcription activators also help recruit the proteins needed to start transcription. These include general transcription factors, RNA polymerase, and co-activators. Together, these proteins form the pre-initiation complex. Activators can bind near a gene promoter or thousands of base pairs away. When they bind far away, DNA bending helps bring them close to the promoter.

These proteins can also help transcription continue if it pauses partway through or needs to start again. Many activators work synergistically, which means several together can produce more transcription than each one alone. Like other proteins, activators can be modified after they are made. For example, acetylation of p53 increases its ability to bind DNA and helps regulate genes involved in tumor suppression.
