Behavioral Video Tracking

Behavioral video tracking is a quantitative method for measuring an organism’s movements and actions from recorded video, converting visual behavior into analyzable data. During analysis, tracking software identifies a subject across successive frames and calculates features such as position, distance traveled, speed, trajectory, time spent in defined regions, or interactions with other subjects. Researchers use these measurements to study locomotion, exploration, social behavior, learning, and responses to environmental or experimental conditions, improving objectivity and reproducibility compared with manual scoring. The approach also supports high-throughput behavioral experiments and enables detailed comparisons across individuals, treatments, and time points.

We have developed a video-rate tracking microscope system that can record and quantify C. elegans behavior at high resolution and high speeds. We have also developed computational methods to reduce the dimensionality of the worm images to a fundamental set of measurements that completely describe the shape of the worm.

Locomotor activity (LMA) is a simple and easily performed measurement of behavior in mice. Coupling of video tracking software (VTS) and LMA allows for the improvement of specificity and sensitivity, especially when compared with the manual, line crossing method of LMA analysis. Additionally VTS allows long-term tracking of mouse LMA.

We describe here a behavioral setup and data analysis method for assaying olfactory responses of up to 100 vinegar flies (Drosophila melanogaster). This system may be used with single or multiple olfactory stimuli, and adaptable for optogenetic activation or silencing of neuronal subsets.
