Four Cups Paradigm

The Four Cups Paradigm is a behavioral experiment that uses four visually similar cups to examine how subjects locate a hidden object or reward, making it useful for studying memory, attention, and decision-making. In a typical trial, a reward is placed beneath one cup while the subject watches, and researchers measure search accuracy, choice latency, and errors as the cup arrangement or visibility of the hiding event changes. By comparing successful retrieval with chance performance, the paradigm helps assess spatial memory, inhibitory control, and the use of visual cues across developmental stages, species, or experimental conditions.

This protocol describes an approach for in toto labeling and multidimensional imaging of zebrafish early eye development. We describe labeling, embedding, and four dimensional (4D) imaging using laser scanning confocal microscopy, and considerations for optimizing acquisition of datasets for dissecting mechanisms of optic cup morphogenesis.

We discussed the synthesis of individual graphitic nanocups using a series of techniques including chemical vapor deposition, acid oxidation and probe-tip sonication. By citrate reduction of HAuCl4, the graphitic nanocups were effectively corked with gold nanoparticles due to the chemically reactive edges of the cups.
