Pilocarpine Injection

Pilocarpine injection is an experimental method that uses a muscarinic acetylcholine receptor agonist to provoke measurable changes in animal behavior and brain activity. After systemic administration, pilocarpine enhances cholinergic signaling, producing progressive behavioral signs such as salivation, tremors, automatisms, and seizures that can develop into status epilepticus, depending on dose and experimental conditions. Researchers use this model to study seizure-related behavior, temporal lobe epilepsy, neuronal injury, and the behavioral effects of candidate anticonvulsant treatments. Careful monitoring of seizure onset, severity, and recovery supports consistent behavioral phenotyping and improves interpretation of neurological outcomes.

This manuscript describes a method of inducing status epilepticus by systemic pilocarpine injection and of monitoring spontaneous recurrent seizures in live animals using a wireless telemetry video and electroencephalogram system. This protocol can be utilized for studying the pathophysiologic mechanisms of chronic epilepsy, epileptogenesis, and acute seizures.
