Shock Tolerance Assessment

Shock tolerance assessment is a behavioral method used to measure how an organism responds to a controlled aversive stimulus and why that response matters for motivation, learning, and welfare research. In a typical assessment, researchers vary the intensity or duration of an electrical shock while recording behaviors such as avoidance, escape, latency, persistence, or willingness to continue a task for a reward. These observations help distinguish sensitivity to aversion from changes in decision-making or performance. In behavior studies, the method can support comparisons across individuals, experimental conditions, or interventions while requiring careful control of stimulus parameters and ethical safeguards.

We describe a protocol to examine the development of opioid-induced hyperalgesia and tolerance in mice. Based on the measurement of thermal and mechanical nociceptive responses of naïve and morphine-treated animals, it allows to quantify the increase in pain sensitivity (hyperalgesia) and decrease in analgesia (tolerance) associated with chronic opiate administration.

The method to measure adult Drosophila associative memory is described. The assay is based on the ability of the fly to associate an odor presented with a negative reinforcer (electric shock) and then recall this information at a later time, allowing memory to be measured.

Bacterial Transformation: The Heat Shock Method

2023

Transformation is the process that occurs when a cell ingests foreign DNA from its surroundings. Transformation can occur in nature in certain types of bacteria. In molecular biology, transformation is artificially reproduced in the lab via the creation of pores in bacterial cell membranes. Bacterial cells that are able to take up DNA from the environment are called competent cells. In the laboratory, bacterial cells can be made competent and DNA subsequently introduced by a procedure called...

This manuscript provides an updated remote supervision protocol that enables participation in transcranial direct current stimulation (tDCS) clinical trials while receiving treatment sessions from home. The protocol has been successfully piloted in both patients with multiple sclerosis and Parkinson's disease.

Salinity Tolerance of Alishewanella Species Isolated from Seawall Muddy Water

Begin with a culture of Alishewanella, a Gram-negative, salt-tolerant bacterium.Next, streak the culture onto agar plates containing increasing concentrations of sodium chloride to simulate varying levels of saline stress.Overlay each plate with a layer of unsolidified medium to prevent oxygen diffusion and maintain the culture in an oxygen-limited environment.Seal and incubate the plates at an optimum temperature to support bacterial growth.During incubation, the bacterium exhibits varying...
