Asymmetric Lipid Composition

Asymmetric lipid composition is the unequal distribution of lipid species between the two leaflets of a biological membrane, a structural feature that influences membrane identity and function. Cells establish and maintain this asymmetry through selective lipid synthesis, transport by flippases, floppases, and scramblases, and differences in leaflet-specific interactions with proteins and the cytoskeleton. In biochemistry, studying lipid asymmetry helps explain membrane curvature, signaling, trafficking, permeability, and recognition of apoptotic cells. Reconstituted membranes and lipidomics allow researchers to examine how composition changes alter membrane organization, providing insight into cellular regulation, disease mechanisms, and the design of biomimetic materials.

Biological membranes show uneven distribution of different types of lipids in the inner and outer layers, resulting in transverse asymmetric membranes. Lipid polyesters constitute the structural components of two cell wall modifications, the plant cuticle and suberin-containing diffusion barriers. In this video, we describe a method to depolymerize cutin from whole delipidated leaves. The method can be applied to investigating mutants compromised in either cutin or suberin biosynthesis.

Here, we present a protocol to quantify precise stepping in rodents. Cortical and the spinal central pattern generator signals are required for precise foot-placement during obstructed locomotion. We report here the novel constrained walking task that directly examines precise stepping behavior.

A protocol is presented to extract the total lipid content of the cell wall of a wide range of mycobacteria. Moreover, extraction and analytical protocols of the different types of mycolic acids are shown. A thin-layer chromatographic protocol to monitor these mycobacterial compounds is also provided.

Experiments on phase separated giant unilamellar vesicles (GUVs) frequently neglect physiological solution conditions. This work presents approaches to study the effect of high-salinity buffer on liquid-liquid phase separation in charged multicomponent GUVs as a function of trans-membrane solution asymmetry and temperature.
