Endosome-enriched Fraction

An endosome-enriched fraction is a biochemical preparation containing a high proportion of endosomes, membrane-bound organelles that sort and transport internalized proteins, lipids, and extracellular material. Researchers typically isolate this fraction through differential centrifugation and density-gradient separation, using differences in organelle size, density, and sedimentation behavior to reduce contamination from other membrane compartments. In biochemistry, endosome-enriched fractions support analysis of cargo trafficking, receptor recycling, endosomal maturation, membrane composition, and protein localization. Although enrichment does not produce a completely pure preparation, it provides a practical material for studying intracellular transport and the molecular mechanisms that regulate endocytic pathways.

Here, we describe a microscopy-based in vitro assay that reconstitutes the nucleation and polymerization of F-actin on early endosomal membranes in test tubes, thus rendering this complex series of reactions amenable to biochemical and genetic manipulations.
