Envelope Stroma

Envelope stroma refers to the stroma-facing compartment associated with the chloroplast envelope, the double-membrane boundary that separates plastid contents from the cytosol. Proteins and metabolites cross the outer and inner envelope membranes through selective transport systems, while the adjacent stroma provides an aqueous environment for enzymes, cofactors, and metabolic reactions. Studying envelope–stroma organization helps explain chloroplast biogenesis, protein targeting, metabolite exchange, and the coordination of processes such as fatty-acid synthesis and photosynthetic carbon assimilation. Biochemical analysis of these compartments also supports research into plant metabolism, organelle communication, and the molecular basis of plastid function.

Outer Layers of the Cell Envelope

2025

The outermost layers of prokaryotic cells play a critical role in their survival, virulence, and interaction with the environment. These layers, often composed of polysaccharides, polypeptides, or proteins, form protective and adhesive structures that vary in organization and function.Capsules and Slime LayersCapsules are highly organized, tightly bound layers that firmly attach to the bacterial cell wall. Capsules are usually made of polysaccharides, though some are made of polypeptides. These...

Protein Transport to the Stroma

2023

Chloroplasts are triple membrane structures with an outer membrane, an inner membrane, and a thylakoid membrane, each containing distinct metabolite transporters, membrane translocons, and enzymes. Appropriate sorting and translocating these proteins to their correct membrane systems is essential for chloroplast function. Protein complexes called the translocon of the outer chloroplast membrane or TOC complex, and the translocon of the inner chloroplast membrane or TIC complex mediate the...

The protocol describes a high-throughput approach to determining structures of membrane proteins using cryo-electron tomography and 3D image processing. It covers the details of specimen preparation, data collection, data processing and interpretation, and concludes with the production of a representative target for the approach, the HIV-1 Envelope glycoprotein. These computational procedures are designed in a way that enables researchers and students to work remotely and contribute to data...

Portal Vein Injection of Colorectal Cancer Organoids to Study the Liver Metastasis Stroma
