Benchtop Transilluminator

A benchtop transilluminator is a compact laboratory instrument that illuminates transparent gels and samples from below to make fluorescent or absorbent bands visible, supporting rapid analysis in biochemistry. It works by directing ultraviolet or blue light through an agarose or polyacrylamide gel; fluorescent stains absorb the excitation light and emit longer-wavelength light that a viewing filter helps isolate, revealing separated nucleic acids or proteins. Researchers use transilluminators to assess electrophoresis results, estimate band size and intensity, document samples, and excise gel bands for downstream experiments. Blue-light models can reduce photodamage and improve handling of sensitive DNA samples.

This protocol demonstrates how to isolate human umbilical cord-derived mesenchymal stem cells' small extracellular vesicles (hUC-MSC-sEVs) with a simple lab-scale benchtop setting. The size distribution, protein concentration, sEVs markers, and morphology of isolated hUC-MSC-sEVs are characterized by nanoparticle tracking analysis, BCA protein assay, western blot, and transmission electron microscope, respectively.
