Functional Activity Assay

A functional activity assay is a laboratory method that measures what a biological molecule does, rather than simply detecting its presence or abundance. In biochemistry, the assay places an enzyme, protein, or other target in controlled conditions and quantifies a functional output, such as substrate conversion, product formation, binding-dependent signaling, or a change in fluorescence or absorbance. Researchers use these measurements to determine activity, compare samples, characterize reaction conditions, evaluate purification and stability, and assess how inhibitors, activators, or mutations affect function. Functional activity assays therefore connect molecular composition with biological performance and support studies of enzyme mechanisms, drug development, and quality control.

Functional Reconstitution and Channel Activity Measurements of Purified Wildtype and Mutant CFTR Protein

Described here is a rapid and effective procedure for functional reconstitution of purified wild-type and mutant CFTR protein that preserves activity for this chloride channel, which is defective in Cystic Fibrosis. Iodide efflux from reconstituted proteoliposomes mediated by CFTR allows studies of channel activity and the effects of small molecules.

This video describes a cell-based reporter assay to determine the estrogenic activity of plant-derived compounds. A compound with estrogen-like activity mimics its binding to estrogen receptors and drives downstream reporter enzyme gene transcription. The reporter enzyme generates bioluminescence in a reaction, indicating the estrogenic activity of the compound.
