Metabolic Alterations Study

A metabolic alterations study investigates changes in metabolites, biochemical pathways, and energy use that occur in cells, tissues, or organisms under specific conditions. Researchers compare metabolic profiles by measuring metabolite concentrations, enzyme activity, or pathway flux before and after a genetic, environmental, or physiological perturbation. These analyses can reveal how altered nutrient utilization, biosynthesis, or energy production reflects cellular responses and disease-related dysfunction. In biochemistry, metabolic alterations studies support the identification of biomarkers, clarification of disease mechanisms, evaluation of drug effects, and development of strategies to restore disrupted metabolic processes.

This protocol describes a method for the large-scale cultivation of Caenorhabditis elegans on solid media. As an alternative to liquid culture, this protocol allows obtaining parameters of different scales under plate-based cultivation. This increases the comparability of results by omitting the morphological and metabolic differences between liquid and solid media culture.

Here we show isolated human platelets can be used as an accessible ex vivo model to study metabolic adaptations in response to the complex I inhibitor rotenone. This approach employs isotopic tracing and relative quantification by liquid chromatography-mass spectrometry and can be applied to a variety of study designs.

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.

What is Metabolism?

2019

Overview Metabolism represents all of the chemical activity in a cell, including reactions that build molecules (anabolism) and those that break molecules down (catabolism). Anabolic reactions require energy, whereas catabolic reactions provide it. Thus, metabolism describes how cells transform energy through a variety of chemical reactions, which are often made more efficient with the help of enzymes. An Introduction to Cell Metabolism

2023

In cells, critical molecules are either built by joining together individual units like amino acids or nucleotides, or broken down into smaller components. Respectively, the reactions responsible for this are referred to as anabolic and catabolic. These reactions require or produce energy typically in the form of a “high-energy” molecule called ATP. Together, these processes make up “Cell Metabolism,” and are hallmarks of healthy, living cells.JoVE’s introduction to cell metabolism briefly...
