One-pot Processing

One-pot processing is a chemical or biochemical method in which multiple sequential operations occur in the same reaction vessel without isolating intermediate products, reducing handling and material loss. Reagents, enzymes, buffers, or other conditions are added in a planned sequence so that one reaction generates the substrate, intermediate, or environment required for the next step, often with controlled changes in pH, temperature, or composition. In biochemistry, this approach can streamline enzyme-catalyzed synthesis, metabolite production, and sample preparation while saving time, solvent, and equipment. It also supports process integration and may improve overall efficiency, although reaction compatibility and intermediate stability must be carefully managed.

2011

A facile, one-pot synthesis of N-succinimidyl-4-[18F]fluorobenzoate ([18F]SFB) was developed based on a non-aqueous, three-step radiochemical process. Using microwave heating, the entire procedure can be completed in less than 30 min, or 60 min with further purification by preparative HPLC. The decay-corrected radiochemical yields (RCYs) were 35-5% (n > 30).

2019

CO2 transformations are conducted in a one-pot two-step procedure for the synthesis of complex molecules. The selective 4 e- reduction of CO2 with a hydroborane reductant affords a reactive and versatile bis(boryl)acetal intermediate which is subsequently involved in condensation reaction or carbene-mediated C-C coupling generation.
