Phosphoproteomics

Phosphoproteomics is the large-scale study of protein phosphorylation, a reversible post-translational modification that regulates protein activity, localization, stability, and interactions. In a typical workflow, proteins are extracted, enzymatically digested into peptides, phosphopeptides are enriched, and mass spectrometry identifies phosphorylation sites and measures their abundance; comparisons across conditions reveal changes in kinase and phosphatase activity. In biochemistry, phosphoproteomics helps map signaling pathways, characterize cellular responses, and identify molecular changes associated with disease or drug treatment. These data provide a systems-level view of phosphorylation-dependent regulation and support the development of biomarkers and targeted therapies.

Proteomic analysis of any cell type is highly dependent on both purity and pre-fractionation of the starting material in order to de-complexify the sample prior to liquid chromatography mass spectrometry (MS). By using back-flushing techniques, pure spermatozoa can be obtained from rodents. Following digestion, phosphopeptides can be enriched using TiO2.
