3d Cell Phenotypes

3D cell phenotypes are the observable structural, functional, and molecular characteristics that cells develop when they grow within three-dimensional environments rather than on flat surfaces. In these models, cell-cell contacts, extracellular-matrix interactions, and gradients of oxygen, nutrients, and signaling molecules shape morphology, organization, gene expression, and behavior. Bioengineers analyze 3D cell phenotypes in spheroids, organoids, hydrogels, and engineered tissues to model development and disease more realistically, evaluate drug responses, and design regenerative therapies. Comparing these phenotypes with conventional two-dimensional cultures can reveal tissue-specific behaviors and improve the predictive value of laboratory models.

Primary cilia influence various signaling pathways. The mammalian cochlea is ideal for examining planar cell polarity (PCP) signaling. Cilia dysfunction affects cochlear outgrowth, cellular patterning and hair cell orientation, readouts of PCP. Our goal is to analyze PCP signaling in mouse cochlea via phenotypic analysis, immunohistochemistry and scanning electron microscopy.

The exposed normal ocular surface consists of cornea and conjunctiva. Epithelial cells, goblet cells and immune cells are present in the conjunctiva. Here, a non-invasive, technique of impression cytology is described using an impression cytology device and flow cytometry to analyze immune cells in the conjunctiva.
