Bead Aggregation Assay

Bead aggregation assay is a method for measuring interactions that cause particles to cluster, providing a practical readout of molecular binding, recognition, or assembly. In a typical design, microspheres are functionalized with specific ligands, antibodies, or biomolecules; interaction with a target connects neighboring beads, producing aggregates that can be quantified through changes in turbidity, light scattering, particle size, or sedimentation. In bioengineering, these assays support biosensor development, evaluation of binding specificity, characterization of engineered biomolecules, and optimization of particle-based systems. Their simple format and adaptable readouts make them useful for screening and studying aggregation-related behavior under controlled conditions.

Bead Aggregation Assays for the Characterization of Putative Cell Adhesion Molecules

Here we present a simple, rapid method for characterizing the intrinsic adhesive properties of putative cell adhesion molecules. The secreted, epitope-tagged ectodomain of a cell adhesion molecule is captured from the culture medium on small, uniform functionalized beads. These beads can then be used immediately in simple bead aggregation assays.

Unmodified and hyperphosphorylated tau proteins were used in two in vitro aggregation assays to reveal the hyperphosphorylation-dependent fast aggregation kinetics. These assays pave the way for future screens for compounds that can modulate the propensity of hyperphosphorylated tau to form fibrils that underlie the progression of Alzheimer’s disease.
