Calcium Dependent Adhesion

Calcium-dependent adhesion is a cell-cell attachment process in which calcium ions regulate molecular interactions, making it important for organizing tissues and designing biomimetic materials. In cadherin-based adhesion, calcium binds extracellular cadherin domains, stabilizing their conformation so proteins on adjacent cells can form selective homophilic bonds; removing calcium weakens or disrupts these contacts. In bioengineering, this mechanism supports the design of cell-adhesive surfaces, engineered tissues, organoid systems, and models of tissue assembly, while calcium control can help tune cell attachment, mechanical cohesion, and remodeling.

Induction of Adhesion-dependent Signals Using Low-intensity Ultrasound

This protocol describes the stimulation of cultured fibroblasts with low-intensity pulsed ultrasound, which drives focal adhesion formation and Rac1 activation by mimicking engagement of the transmembrane matrix receptor, syndecan-4. This approach allows investigation of a successful clinical technique at the cellular level, thereby providing opportunities for refinement of the therapy.

Calcium-Dependent Hydrophobic Interaction Chromatography: A Technique to Purify Calcium-Binding Proteins Based on Hydrophobic Interactions

In this video, we demonstrate the purification of calcium-binding protein from a dialyzed cell lysate through calcium-dependent hydrophobic interaction chromatography. The calcium-binding proteins expose a hydrophobic region upon binding with calcium, facilitating interaction with a hydrophobic group on resin. Later these proteins are eluted using calcium chelator EDTA that reverses the interaction.
