Cell-free Protein Expression

Cell-free protein expression is the production of proteins outside living cells using biological molecular machinery in a controlled reaction mixture. A DNA or RNA template directs transcription and translation in a cell lysate or reconstituted system supplied with ribosomes, enzymes, amino acids, energy, and cofactors. By eliminating cell growth and membrane barriers, the method enables rapid adjustment of reaction conditions and expression of proteins that may be toxic or difficult to produce in living organisms. In bioengineering, cell-free systems support genetic circuit prototyping, biosensor development, enzyme synthesis, therapeutic protein research, and on-demand manufacturing.

Expression of malarial proteins in cell based systems remains challenging. We demonstrate two step and one step IVT (in vitro translation) cell free expression systems for expressing malarial recombinant rhoptry proteins from HeLa cells. We use a Ni-resin affinity based purification system to purify the rhoptry proteins.

In the post-human genomics era, the availability of recombinant proteins in native conformations is crucial to structural, functional and therapeutic research and development. Here, we describe a test- and large-scale protein expression system in human embryonic kidney 293T cells that can be used to produce a variety of recombinant proteins.
