Collagen Matrix Encapsulation

Collagen matrix encapsulation is a bioengineering technique that embeds cells, biomolecules, or engineered tissues within a three-dimensional collagen scaffold to reproduce aspects of the extracellular matrix. During controlled gelation, collagen molecules assemble into a fibrillar network that physically retains the encapsulated material while providing sites for cell adhesion, signaling, and transport of nutrients and waste. This tunable microenvironment supports studies of cell behavior, tissue formation, and disease processes under more physiologically relevant conditions than two-dimensional culture. Applications include tissue engineering, regenerative medicine, organoid models, biomaterial development, and in vitro evaluation of therapeutics.

Cell migration is an important part of human development and life. In order to understand the mechanisms that can alter cell migration, we present a planar gradient diffusion system to investigate chemotaxis in a 3D collagen matrix, which allows one to overcome modern diffusion chamber limitations of existing assays.

Dissection and Culture of Mouse Dopaminergic and Striatal Explants in Three-Dimensional Collagen Matrix Assays

Explants from the midbrain dopamine system and striatum are used in a collagen matrix assay for the in vitro analysis of mesostriatal and striatonigral pathway development. In this assay axonal outgrowth and guidance can be manipulated and quantified. It can also be modified for assessing other regions or molecular cues.
