Cytochrome Absorbance

Cytochrome absorbance is the measurement of light absorbed by cytochromes, heme-containing proteins that participate in electron transfer and cellular energy conversion. Their heme groups produce characteristic absorption bands, and changes in absorbance occur when the iron center shifts between oxidized and reduced states, allowing spectrophotometric monitoring of redox reactions. In bioengineering, absorbance analysis helps characterize electron-transport pathways, assess mitochondrial or microbial respiration, and evaluate engineered cells, enzymes, and biocatalytic systems. These measurements can also support biosensor development and process monitoring by linking optical signals to biochemical activity and oxygen or electron-acceptor utilization.

In this protocol, we describe the direct cytoplasmic microinjection of cytochrome c protein into fibroblasts and primary sympathetic neurons. This technique allows for the introduction of cytochrome c protein into the cytoplasm of cells and mimics the release of cytochrome c from mitochondria, which occurs during apoptosis.

This procedure describes how to encapsulate cytochrome c (cyt. c) in silica (SiO2) sol-gels, process these gels to form bioaerogels, and use these bioaerogels to rapidly recognize nitric oxide (NO) through a gas-phase reaction. This type of protocol may aid in the future development of biosensors or other bioanalytical devices.

Solution-suspendable gold nanotubes with controlled dimensions can be synthesized by electrochemical deposition in porous anodic aluminum oxide (AAO) membranes using a hydrophobic polymer core. Gold nanotubes and nanotube arrays hold promise for applications in plasmonic biosensing, surface-enhanced Raman spectroscopy, photo-thermal heating, ionic and molecular transport, microfluidics, catalysis and electrochemical sensing.
