Single Crrna Array

A single crRNA array is a engineered CRISPR RNA construct that encodes one or more guide sequences in a unified transcript, enabling programmable nucleic acid targeting. After transcription, system-specific processing enzymes cleave the precursor at direct-repeat regions to release mature CRISPR RNAs, which pair with a Cas protein and guide it to complementary DNA or RNA sequences through base pairing. In bioengineering, this design supports multiplex genome editing, transcriptional regulation, and targeted screening by coordinating several guides from one genetic cassette. Its compact organization can simplify vector construction and improve the control of complex cellular engineering workflows.

Implanting organized arrays of microwires for use in single-unit electrophysiological recordings presents a number of technical challenges. Methods for performing this technique and the equipment necessary are described. Also, the beneficial use of organized microwire arrays to record from distinct neural subregions with high spatial selectivity is discussed.

Design and Use of Multiplexed Chemostat Arrays

We developed and validated a small-footprint array of miniature chemostats built from readily available parts for low cost. Physiological and experimental evolution results were similar to larger volume chemostats. The ministat array provides a compact, inexpensive, and accessible platform for traditional chemostat experiments, functional genomics, and chemical screening applications.

Described are protocols for quantifying mechanical interactions between adherent cells and microstructured substrates. These interactions are closely linked to essential cell behaviors including migration, proliferation, differentiation, and apoptosis. The protocols present an open-source image analysis software called MechProfiler, which enables determination of involved forces for each micropost.
